Cytokines and growth factors are traditionally expressed in bacteria or insect cells. Those recombinant proteins have been widely used in cell biology, immunology and stem cell research. However, there are many drawbacks associated with bacteria and insect cells-generated cytokines, such as endotoxin contamination, incorrect conformation, no or aberrant glycosylation, low biological activity and instability. New technologies developed in recent years have finally made it possible to cost effectively manufacture authentic human cytokines with natural conformation and native glycosylation. The HumanKine™ cytokines listed below are a group of premium human cytokines that have been produced with our proprietary technologies, which renders unprecedented advantages over other cytokines on the market:

Endotoxin-free

The cytokines are produced from human cells under strict endotoxin-free conditions. No endotoxin was ever detected in our products to the sensitivity limit of several commercial endotoxin detection kits. Human cytokines are frequently used in immunology research. Endotoxin (LPS) is very powerful in activating many types of immune cells (such as dendritic cells and macrophages) and triggering immune responses. Trace amount of LPS contamination in cytokines may obscure the true phenomenon under study. Using LPS-free cytokines is essential to most immunology experiments, especially for in vitro differentiation of many cell types (Th1, Th2, Th17, Treg, DC, etc) by long-term incubation with cytokines.

Natural structure and natural glycosylation

The cytokines are produced from stable HEK293 cell lines. Recombinant proteins expressed by this human cell line are most similar (if not identical) to natural proteins in terms of glycosylation pattern and conformation. Proteins produced from E. coli, Yeast, Incest cells and even CHO cells often have no glycosylation or aberrant glycosylation, incorrect structure, mis-paired disulfide bonds and mis-organized multimerizations. Human cells are the best choice in producing human proteins.

High activity

The cytokines are usually more active than those from other sources, largely due to correct conformation and post-translation modifications.

High stability

Their natural conformation and post-translation modifications also translate into longer half-life in vivo and in cell cultures.

Negligible batch-to-batch variation

The cytokines are produced from Stably-transfected HEK293 cell lines. Batch-to-batch variation is minor when compared to those produced by transient expressions.

Contaminants-free

The cell lines are cultured in chemically defined serum-free medium, without any components of animal source. Contamination of animal-borne viruses and other agents are near impossible.

Unlimited supply

The cell lines stably secrete target cytokines continuously for more than 20 passages without any decline in expression level. Their production potential is virtually unlimited.

Easy to convert to human use

The cell lines are scalable to large bioreactors. Our cell lines are stable, free of any viruses and already adapted to chemically defined medium. Should clinical trials are needed with the advance of preclinical researches, large quantity of clinical grade therapeutic proteins can be easily obtained to meet regulatory, safety, and consistency requirements.
