GelSafe™ is a new and acid stain for double-stranded DNA, single-stranded DNA, and RNA in agarose gels. The dyes are developed to replace toxic Ethidium Bromide (EB, a potent mutagen), commonly used in gel electrophoresis for visualization of nucleic acids in agarose gels. GelSafe™ is non-carcinogenic by Ames-test. Results are negative in both mouse marrow chromophilous erythrocyte micronucleus and mouse spermary spermatocyte chromosomal aberration tests.

1 ml (20,000X, use 5µl for 100ml of gel and gel running buffer)

Usage

GelSafe™ is used the same way as Ethidium Bromide in agarose gel electrophoresis. It emits green fluorescence when bound to dsDNA and red fluorescence when bound to ssDNA or RNA. This stain has two fluorescence excitation maxima when bound to nucleic acid, at approximately 290nm and 490nm.
It is recommended to add GelSafe™ to running buffer as well (although it is not always required), and view the results under standard UV light.

Sensitivity

0.1-0.3 ng/band (Ethidium Bromide’s sensitivity is 1-5 ng/band)
Note: In gel documentation systems optimized for EtBr, GelSafe™ may appear less bright than EtBr.

Disposal   

Dispose GelSafe™ as you would for any other non-carcinogenic fluorescent dye (eg. Acridine orange; Propidium iodide).
