CTLA-4 (CD152) is a cell receptor constitutively expressed on Treg cells and activated T cells. When bound to CD80 or CD86 on antigen presenting cells, the molecule exerts inhibitory functions to activated T cell, thus acts as a "break" for immune responses. Using CTLA-4 mimics can reduce over response of immune system to self antigen, thus is useful for treating autoimmune diseases such as rheumatoid arthritis. Blocking CTLA-4 with monoclonal antibodies can enhance immune responses to tumors. Several drugs targeting to CTLA-4 have been approved or in late clinical stage, including Abatacept (Orencia), ipilimumab (Yervoy) and tremelimumab.
