Programmed cell death protein 1 (PD-1), also known as CD279, is a transmembrane protein that is expressed on the surface of activated T cells, B cells, and macrophages. The molecule negatively regulates T cell activation and serves as a “brake” of immune responses against tumors and infectious diseases. Blocking PD-1 with monoclonal antibodies can lift the “brake” and make cancer cells susceptible to attacks by T cells. A few such anti-PD-1 monoclonal antibodies have been approved by FDA for treatment of various cancers, including nivolumab (Opdivo by Bristol Myers Squibb) and Pembrolizumab (Keytruda by Merck).
