Cell membrane coated nanoparticles (MNPs) harboring target full-length multi-transmembrane protein in its native form were produced by overexpression of the protein in HEK293 cells followed by membrane extraction and extrusion. The target transmembrane protein maintains its natural conformation embedded in natural cell membrane coated on nanoparticles.
This specific membrane protein, the human full length TSPAN33 protein (Uniprot ID: Q86UF1), has a MW of 31.5 kDa.

Elisa plates were pre-coated with 0.5ug/well purified human TSPAN33 full length membrane nanoparticles. Serial diluted anti-Flag monoclonal antibody solutions were added, washed, and incubated with secondary antibody before Elisa reading. Calculated from the data, EC50 for anti-Flag monoclonal antibody binding with TSPAN33 full length membrane nanoparticles is 13.66ng/ml.
