Synthetic nanodiscs are produced by solubilizing HEK293 cell membrane fractions overexpessing target full-length multi-transmembrane protein with synthetic polymers such as styrene-maleic acid co-polymers (SMAs), forming discoidal nanostructures with target protein intact in lipid bilayer. The nanodiscs are further purified, offering intact membrane proteins in their natural configuration. This human full length CCR8 Protein has a MW of 40.7 kDa.

formulation Lyophilized from nanodisc solubilization buffer (20 mM Tris-HCl, 150 mM NaCl, pH 8.0). Normally 5% - 8% trehalose is added as protectants before lyophilization.
reconstitution Reconstitute lyophilized powder with water to a concentration of no less than 100 ug/ml, solubilize at RT for 30 min with occasional gentle mixing. It is recommended that 0.1% of HSA be added as a carrier protein for long term storage. Do not use solvents with pH lower than 6.5 in subsequent experiments.

storage Lyophilized form is stable for 1 week at room temperature. Store at -20°C to -80°C for 12 months in lyophilized form. After reconstitution, store at 4°C for 2-7 days or at below -20°C for up to 3 months.

Elisa plates were pre-coated with Flag Tag CCR8-Nanodisc (0.2 ug per well ). Serial diluted anti-CCR8 monoclonal antibodysolutions were added, washed, and incubated with secondary antibody before Elisa reading. From above data, the EC50 for anti-CCR8 monoclonal antibody binding with CCR8-Nanodisc is 12.07 ng/ml.
