Synthetic nanodiscs are produced by solubilizing HEK293 cell membrane fractions overexpessing target full-length multi-transmembrane protein with synthetic polymers such as styrene-maleic acid co-polymers (SMAs), forming discoidal nanostructures with target protein intact in lipid bilayer. The nanodiscs are further purified, offering intact membrane proteins in their natural configuration. This human full length S1PR1-Strep protein has a MW of 42.8 kDa.

Elisa plates were pre-coated with C-Flag&Strep Tag S1PR1-Strep-Nanodisc (0.2 ug/per well). Serial diluted anti-Flag monoclonal antibody solutions were added, washed, and incubated with secondary antibody before Elisa reading. From above data, the EC50 for anti-Flag monoclonal antibody binding with S1PR1-Strep-nanodisc is 4.824 ng/mL.
