Immunohistochemistry: IHC-P analysis of uterus tissue by FGF2 antibody (KT49). IHC-P was performed using sections of the formalin-fixed paraffin-embedded uterus tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug KT49 with SK-OV-3 lysate containing 200 ug total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto PVDF membrane. FGF2 (KT49) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG (Fc specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: SK-OV-3 lysate Lane 2: FGF2 immunoprecipitated from SK-OV-3 lysate by KT49 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: KT49 can immunoprecipitate FGF2.
Western Blot: 15 ug of SK-OV-3 lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT49 at 1 ug/mL was used as the primary antibody and peroxidase-conjugated goat anti-mouse IgG was used as the secondary antibody. FGF2 band was visualized using ECL Substrate. Result: KT49 can detect FGF2 by Western blotting.
