Immunohistochemistry: IHC-P analysis of testis tissue by MDM2 antibody (K56006_10C9). IHC-P was performed using sections of the formalin-fixed paraffin-embedded testis tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K56006_10C9 with 200 ug of cell lysate extracted from Nutlin3a treated A549. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. MDM2 (86934S) with a dilution ratio of 1:1000 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: A549 lysate (treated with 10 uM Nutlin3a for 24 h) Lane 2: MDM2 immunoprecipitated by K56006_10C9 from the same lysate as Lane 1 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K56006_10C9 can immunoprecipitate MDM2.
