ELISA: Microtiter wells were coated with K94024_4A9 at 5 ug/mL as the capture antibody. Recommbinant HSPB1 Protein (Cat. PP-086) was used as the antigen. 1 ug/mL biotin conjugated HSPB1 Antibody (K1E017_2G11) was used as the detection antibody. SA-HRP was then added to the system. Result: K94024_4A9 and K1E017_2G11 can be used as a matched antibody pair to detect and quantify the concentration of HSPB1 .
Immunohistochemistry: IHC-P analysis of heart muscle tissue by HSPB1 antibody (K94024_4A9). IHC-P was performed using sections of the formalin-fixed paraffin-embedded heart muscle tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K94024_4A9 with HeLa lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. HSPB1 (K1E017_3H10) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG (Fc specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: HeLa lysate Lane 2: HSPB1 immunoprecipitated from HeLa lysate by K94024_4A9 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K94024_4A9 can immunoprecipitate HSPB1.
Western Blot: 15 ug of THP-1 lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K94024_4A9 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. HSPB1 band was visualized using ECL Western Blotting Substrate. Result: K94024_4A9 can detect HSPB1 by Western blotting.
