Immunohistochemistry: IHC-P analysis of tonsil tissue by BCL6 antibody (K1E024_3B12). IHC-P was performed using sections of the formalin-fixed paraffin-embedded tonsil tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K1E024_3B12 with Ramos lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. BCL6 (K1E024_3B12) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: Ramos lysate Lane 2: BCL6 immunoprecipitated from Ramos lysate by K1E024_3B12 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K1E024_3B12 can immunoprecipitate BCL6.
Western Blot: 15 ug of Ramos lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K1E024_3B12 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. BCL6 band was visualized using ECL Western Blotting Substrate. Result: K1E024_3B12 can detect BCL6 by Western blotting.
