Immunohistochemistry: IHC-P analysis of colon tissue by TGM2 antibody (K29035_9F5). IHC-P was performed using sections of the formalin-fixed paraffin-embedded lung cancer tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K29035_9F5 with A549 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. TGM2 (K29035_14E8) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: A549 lysate Lane 2: TGM2 immunoprecipitated from A549 lysate by K29035_9F5 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K29035_9F5 can immunoprecipitate TGM2.
Western Blot: 15 ug of A549 lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K29035_9F5 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. TGM2 band was visualized using ECL Western Blotting Substrate. Result: K29035_9F5 can detect TGM2 by Western blotting.
