Immunohistochemistry: IHC-P analysis of esophageal cancer tissue by ARID1A antibody (KAA140_16E7). IHC-P was performed using sections of the formalin-fixed paraffin-embedded esophageal cancer tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of KAA140_16E7 with Jurkat lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. ARID1A (KAA140_16E7) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: Jurkat lysate Lane 2: ARID1A immunoprecipitated from Jurkat lysate by KAA140_16E7 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: KAA140_16E7 can immunoprecipitate ARID1A.
Western Blot: 15 ug of Jurkat lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KAA140_16E7 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. ARID1A band was visualized using ECL Western Blotting Substrate. Result: KAA140_16E7 can detect ARID1A by Western blotting.
