Immunohistochemistry: IHC-P analysis of tonsil tissue by CR2 antibody (KAA120_10G5). IHC-P was performed using sections of the formalin-fixed paraffin-embedded tonsil tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug KAA120_10G5 with Raji lysate containing 200 ug total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. CR2 (KAA120_10G5) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: 7.5 ug of Raji lysate Lane 2: CR2 immunoprecipitated from Raji lysate by KAA120_10G5 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: KAA120_10G5 can immunoprecipitate CR2.
Western Blot: 15 ug of Raji lysate was run SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KAA120_10G5 at 1 ug/mL was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. CR2 band was visualized using ECL Substrate. Result: KAA120_10G5 can detect CR2 by Western blotting.
