Immunohistochemistry: IHC-P analysis of brain tissue by CALB2 antibody (KAA343_8F8). IHC-P was performed using sections of the formalin-fixed paraffin-embedded brain tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug KAA343_8F8 with rat brain tissue lysate containing 200 ug total protein. After absorption with Protein G beads the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. Anti-CALB2 (KAA343_8F8) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG (Fc specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: Rat brain tissue lysate Lane 2: CALB2 immunoprecipitated from rat brain tissue lysate by KAA343_8F8 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: KAA343_8F8 can immunoprecipitate CALB2.
Western Blot: 15 ug of rat brain tissue lysate and 15 ug of mouse brain tissue lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KAA343_8F8 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. CALB2 band was visualized using ECL Western Blotting Substrate. Lane 1: 15 ug of rat brain tissue lysate Lane 2: 15 ug of mouse brain tissue lysate Result: KAA343_8F8 can detect CALB2 by Western blotting.
