Immunohistochemistry: IHC-P analysis of malignant melanoma tissue by MLANA antibody (KAA343_6B7). IHC-P was performed using sections of the formalin-fixed paraffin-embedded malignant melanoma tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of KAA343_6B7 with SK-MEL-28 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. MLANA (K16232_11F5) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG (Fc specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: SK-MEL-28 lysate Lane 2: MLANA immunoprecipitated from SK-MEL-28 lysate by KAA343_6B7 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: KAA343_6B7 can immunoprecipitate MLANA.
Western Blot: 15 ug of SK-MEL-28 lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KAA343_6B7 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. MLANA band was visualized using ECL Western Blotting Substrate. Result: KAA343_6B7 can detect MLANA by Western blotting.
