Immunohistochemistry: IHC-P analysis of lung tissue by MSR1 antibody (K52021_3G12). IHC-P was performed using sections of the formalin-fixed paraffin-embedded lung tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K52021_3G12 with PMA treated THP-1 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. MSR1 (K52021_3G12) at 1 ug/mL was used as the primary antibody and peroxidase conjugated rabbit anti-mouse IgG (Light chain specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: THP-1 treated with 20 ng/mL PMA Lane 2: MSR1 immunoprecipitated from THP-1 treated with 20 ng/mL PMA by K52021_3G12 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K52021_3G12 can immunoprecipitate MSR1.
Western Blot: THP-1 cells were treated with 20ng/mL PMA. 15 ug of the cell lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K52021_3G12 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. MSR1 band was visualized using ECL Substrate. Result: K52021_3G12 can detect MSR1 by Western blotting.
