Immunocytochemistry analysis of SQSTM1/p62 (green) in U87-MG using SQSTM1/p62 antibody,and DAPI(blue).
Immunohistochemistry: IHC-P analysis of breast cancer tissue by SQSTM1 antibody. IHC-P was performed using sections of the formalin-fixed paraffin-embedded breast cancer tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 10 uL of RC-3039 with HeLa lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. SQSTM1 (RC-3039) with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was Rabbit IgG. Lane 1: HeLa lysate Lane 2: SQSTM1 immunoprecipitated from HeLa lysate by RC-3039 Lane 3: The same as Lane 2 but Rabbit IgG was used as IgG isotype control antibody Result: RC-3039 can immunoprecipitate SQSTM1.
Western Blot: 15 ug of K-562 lysate and 15 ug of HeLa lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. RC-3039 with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. SQSTM1 band was visualized using ECL Western Blotting Substrate. Lane 1: 15 ug of K-562 lysate Lane 2: 15 ug of HeLa lysate Result: RC-3039 can detect SQSTM1 by Western blotting.
