Immunoprecipitation: Immunoprecipitation was performed by incubation of 10 uL of RC-3048 with A549 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. OLIG1 (RC-3048) with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was Rabbit IgG. Lane 1: A549 lysate Lane 2: OLIG1 immunoprecipitated from A549 lysate by RC-3048 Lane 3: The same as Lane 2 but Rabbit IgG was used as IgG isotype control antibody Result: RC-3048 can immunoprecipitate OLIG1.
Western Blot: Various protein samples were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. RC-3048 with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. OLIG1 band was visualized using ECL Western Blotting Substrate. Lane 1: 15 ug of NIH/3T3 lysate Lane 2: 15 ug of A569 lysate Lane 3: 15 ug of HL-60 lysate Lane 4: 15 ug of U-2 OS lysate Result: RC-3048 can detect OLIG1 by Western blotting.
