Immunoprecipitation: Immunoprecipitation was performed by incubation of 10 uL of RC-3060 with rat brain tissue lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. PPP3CA (RC-3060) with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was Rabbit IgG. Lane 1: Rat brain tissue lysate Lane 2: PPP3CA immunoprecipitated from rat brain tissue lysate by RC-3060 Lane 3: The same as Lane 2 but Rabbit IgG was used as IgG isotype control antibody Result: RC-3060 can immunoprecipitate PPP3CA.
Western Blot: 15 ug of U-2 OS lysate and 15 ug of rat brain tissue lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. RC-3060 with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. PPP3CA band was visualized using ECL Western Blotting Substrate. Lane 1: 15 ug of U-2 OS lysate Lane 2: 15 ug of rat brain tissue lysate Result: RC-3060 can detect PPP3CA by Western blotting.
