Immunocytochemistry analysis of Gemin 1(green) in Hela using Gemin 1 antibody,and DAPI(blue)
Immunoprecipitation: Immunoprecipitation was performed by incubation of 10 uL of RC-3061 with HeLa lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. SMN1 (RC-3061) with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was Rabbit IgG. Lane 1: HeLa lysate Lane 2: SMN1 immunoprecipitated from HeLa lysate by RC-3061 Lane 3: The same as Lane 2 but Rabbit IgG was used as IgG isotype control antibody Result: RC-3061 can immunoprecipitate SMN1.
Western Blot: 15 ug of K-562 lysate and 15 ug of HeLa lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. 1Z572024-L24 with a dilution ratio of 1:750 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. SMN band was visualized using ECL Western Blotting Substrate. Lane 1: 15 ug of K-562 lysate Lane 2: 15 ug of HeLa lysate Result: 1Z572024-L24 can detect SMN by Western blotting.
