Immunohistochemistry: IHC-P analysis of lung tissue by ICAM1 antibody. IHC-P was performed using sections of the formalin-fixed paraffin-embedded lung tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 10 uL of RC-3738 with Raji lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. ICAM1 (RC-3738) with a dilution ratio of 1:1000 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was Rabbit IgG. Lane 1: Raji lysate Lane 2: ICAM1 immunoprecipitated from Raji lysate by RC-3738 Lane 3: The same as Lane 2 but Rabbit IgG was used as IgG isotype control antibody Result: RC-3738 can immunoprecipitate ICAM1.
Western Blot: 15 ug of Raji lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. RC-3738 with a dilution ratio of 1:1000 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. ICAM1 band was visualized using ECL Western Blotting Substrate. Result: RC-3738 can detect ICAM1 by Western blotting.
