Immunoprecipitation: Immunoprecipitation was performed by incubation of 10 uL of RC-3944 with K-562 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. CD71 (RC-3944) with a dilution ratio of 1:1000 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. The isotype control antibody was Rabbit IgG. Lane 1: K-562 lysate Lane 2: CD71 immunoprecipitated from K-562 lysate by RC-3944 Lane 3: The same as Lane 2 but Rabbit IgG was used as IgG isotype control antibody Result: RC-3944 can immunoprecipitate CD71.
Western Blot: 15 ug of HeLa lysate and 15 ug of K-562 lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. RC-3944 with a dilution ratio of 1:1000 was used as the primary antibody and peroxidase conjugated goat anti-rabbit IgG was used as the secondary antibody. CD71 band was visualized using ECL Western Blotting Substrate. Lane 1: 15 ug of HeLa lysate Lane 2: 15 ug of K-562 lysate Result: RC-3944 can detect CD71 by Western blotting.
