Synthetic nanodiscs are produced by solubilizing HEK293 cell membrane fractions overexpessing target full-length multi-transmembrane protein with synthetic polymers such as styrene-maleic acid co-polymers (SMAs), forming discoidal nanostructures with target protein intact in lipid bilayer. The nanodiscs are further purified, offering intact membrane proteins in their natural configuration. This human full length TSHR protein has a MW of 86.8 kDa.

Elisa plates were pre-coated with Flag Tag TSHR-Nanodisc (0.2 ug per well ). Serial diluted anti-Flag monoclonal antibody solutions were added, washed, and incubated with secondary antibody before Elisa reading. From above data, the EC50 for anti-Flag monoclonal antibody binding with TSHR-Nanodisc is 2.108 ng/ml.
