Cross reactivity test: Microtiter wells were coated with various human cytokines. K29001_11F4 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: K29001_11F4 does not cross-react with human IL1B, IL2, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, CSF2, IFNA, CCL2, KITLG and TNF.
ELISA: Microtiter wells were coated with K29001_11F4 at 3 ug/mL as the capture antibody. Recombinant human IFNG (Cat. PP-486) was used as the antigen. Peroxidase conjugated mouse anti-human IFNG monoclonal antibody (KT218) was used as the detection antibody. Result: K29001_11F4 and KT218 can be used as a matched antibody pair to detect and quantify the concentration of IFNG.
Western Blot: 25ng of recombinant human IFNG was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K29001_11F4 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. IFNG band was visualized using ECL Substrate. Result: K29001_11F4 can detect recombinant human IFNG by Western blotting.
