Cross reactivity test: Microtiter wells were coated with various human cytokines. KT218 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: KT218 does not cross-react with human IL1B, IL2, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, CSF2, IFNA, CCL2, KITLG and TNF.
ELISA: Microtiter wells were coated with KT218 at 3 ug/mL as the capture antibody. Human IFNG was used as the antigen. Peroxidase conjugated mouse anti-human IFNG monoclonal antibody (K29001_11F4) was used as the detection antibody. Result: KT218 and K29001_11F4 can be used as a matched antibody pair to detect and quantify the concentration of IFNG.
Western Blot: 100 ng of recombinant human IFNG was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT218 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. IFNG band was visualized using ECL Substrate. Result: KT218 can detect recombinant human IFNG by Western blotting.
