Cross reactivity test: Microtiter wells were coated with human immunoglobulins with various isotypes. KT16 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: KT16 does not cross-react with human IgG1, IgG2, IgG3, IgG4, IgA and IgE

ELISA: Microtiter wells were coated with KT16 at 5.0 ug/mL as the capture antibody. IGHM (Cat. KC-007) was used as the antigen. Peroxidase conjugated mouse anti-human IGHM monoclonal antibody (KT38) was used as the detection antibody. Result: KT16 and KT38 can be used as a matched antibody pair to detect and quantify the concentration of IGHM .

Western Blot: Human IgG, human IgA, human IgM and human IgE were run on 6-18% SDS-PAGE under reducing conditions and blotted onto PVDF membrane. KT16 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Human IgM band was visualized using ECL Western Blotting Substrate. Lane 1: 100 ng of human IgG Lane 2: 100 ng of human IgA Lane 3: 100 ng of human IgM Lane 4: 100 ng of human IgE Result: KT16 can specifically detect human IgM by Western blotting.
