Immunohistochemistry: IHC-P analysis of spleen tissue by CDA antibody (K1E006_4H7). IHC-P was performed using sections of the formalin-fixed paraffin-embedded spleen tissue.
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug K1E006_4H7 with SK-OV-3 lysate containing 200 ug total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. CDK4 (K1E006_15H12) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: SK-OV-3 lysate Lane 2: CDK4 immunoprecipitated from SK-OV-3 lysate by K1E006_4H7 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K1E006_4H7 can immunoprecipitate CDK4.
Western Blot: 30 ug of HeLa lysate and 30 ug of SK-OV-3 lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K1E006_4H7 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. CDA band was visualized using ECL Substrate. Lane 1: 30 ug of HeLa lysate Lane 2: 0 ug of SK-OV-3 lysate Result: K1E006_4H7 can detect CDA by Western blotting.
