ELISA: Microtiter wells were coated with K52006_16F8 at 3.0 ug/mL as the capture antibody. MMP9 (Cat. PQ-345) was used as the antigen. Peroxidase conjugated mouse anti-human MMP9 monoclonal antibody (K52006_14E11) was used as the detection antibody. Result: K52006_14E11 and K52006_16F8 can be used as a matched antibody pair to detect and quantify the concentration of MMP9 .
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K52006_14E11 with PMA treated U-937 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. MMP9 (K52006_2E9) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: PMA treated U-937 lysate Lane 2: MMP9 immunoprecipitated from PMA treated U-937 lysate by K52006_14E11 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K52006_14E11 can immunoprecipitate MMP9.
Western Blot: 15 ug of PMA treated U-937 lysate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K52006_14E11 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. MMP9 band was visualized using ECL Western Blotting Substrate. Result: K52006_14E11 can detect MMP9 by Western blotting.
