Immunoprecipitation: Immunoprecipitation was performed by incubation of 5 ug KT142 with rat heart lysate containing 500 ug total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto PVDF membrane. Anti-TNNI3 (KT142) at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG (Fc specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: Rat heart lysate Lane 2: TNNI3 immunoprecipitated from rat heart lysate by KT142 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: KT142 can immunoprecipitate TNNI3.
Western Blot: 15 ug of rat heart lysate were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT142 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. TNNI3 band was visualized using ECL Substrate. Result: KT142 can detect TNNI3 by Western blotting.
