ELISA: Microtiter wells were coated with K70050_11B4 at 3.0 ug/mL as the capture antibody. SIGLEC7 (Cat. PP-1188) was used as the antigen. Peroxidase conjugated mouse anti-human SIGLEC7 monoclonal antibody (K70050_6H4) was used as the detection antibody. Result: K70050_11B4 and K70050_6H4 can be used as a matched antibody pair to detect and quantify the concentration of SIGLEC7 .
Immunoprecipitation: Immunoprecipitation was performed by incubation of 2.5 ug of K70050_11B4 with HL-60 lysate containing 200 ug of total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. SIGLEC7 (OC-104) at 1 ug/mL was used as the primary antibody and peroxidase conjugated rabbit anti-mouse IgG (Light chain specific) was used as the secondary antibody. The isotype control antibody was KT82. Lane 1: HL-60 lysate Lane 2: SIGLEC7 immunoprecipitated from HL-60 lysate by K70050_11B4 Lane 3: The same as Lane 2 but KT82 was used as IgG isotype control antibody Result: K70050_11B4 can immunoprecipitate SIGLEC7.
