Cross reactivity test: Microtiter wells were coated with various human cytokines. K06301_19H9 was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: K06301_19H9 does not cross-react with human IL1B, IL2, IL4, IL5, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, CSF2, IFNA, IFNG, CCL2, KITLG and TNF.
ELISA: Microtiter wells were coated with K06301_19H9 at 3.0 ug/mL as the capture antibody. IL6 (Cat. PP-496) was used as the antigen. Peroxidase conjugated mouse anti-human IL6 monoclonal antibody (K06301_22C3) was used as the detection antibody. Result: K06301_19H9 and K06301_22C3 can be used as a matched antibody pair to detect and quantify the concentration of IL6 .
Western Blot: 25 ng of recombinant human IL-6 was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K06301_19H9 at 1 ug/mL was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. IL-6 band was visualized using ECL Substrate. Result: K06301_19H9 can detect recombinant human IL-6 by Western blotting
