Cross reactivity test: Microtiter wells were coated with various human ytokines. KT221 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: KT221 does not cross-react with human IL1B, IL2, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, CSF2, IFNA, IFNG, KITLG and TNF.
ELISA: Microtiter wells were coated with KT221 at 3.0 ug/mL as the capture antibody. CCL2 (Cat. PP-538) was used as the antigen. Peroxidase conjugated mouse anti-human CCL2 monoclonal antibody (K06288_4G5) was used as the detection antibody. Result: KT221 and K06288_4G5 can be used as a matched antibody pair to detect and quantify the concentration of CCL2 .
Western Blot: 25 ng of recombinant human CCL2 was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT221 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. CCL2 band was visualized using ECL Substrate. Result: KT221 can detect recombinant human CCL2 by Western blotting.
