Cross reactivity test: Microtiter wells were coated with various human cytokines. K06288_1B2 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: K06288_1B2 does not cross-react with human IL1B, IL2, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, CSF2, IFNA, IFNG, CCL2 and KITLG.
ELISA: Microtiter wells were coated with K06288_1B2 at 3.0 ug/mL as the capture antibody. TNF (Cat. PQ-342) was used as the antigen. Peroxidase conjugated mouse anti-human TNF monoclonal antibody (K03008_3A9) was used as the detection antibody. Result: K06288_1B2 and K03008_3A9 can be used as a matched antibody pair to detect and quantify the concentration of TNF .
Western Blot: 25 ng of recombinant human TNF was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K06288_1B2 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. TNF band was visualized using ECL Substrate. Result: K06288_1B2 can detect recombinant human TNF by Western blotting.
