Cross reactivity test: Microtiter wells were coated with various human cytokines. KT31 was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: KT31 does not cross-react with human IL1B, IL2, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, IL17A, CSF2, IFNA, IFNG, CCL2 and KITLG.

ELISA: Microtiter wells were coated with KT31 at 3 ug/mL as the capture antibody. Human TNF was used as the antigen. Peroxidase conjugated mouse anti-human TNF (K06288_1B2) was used as the detection antibody. Result: KT31 and K06288_1B2 can be used as a matched antibody pair to detect and quantify the concentration of human TNF.

Western Blot: 25 ng of recombinant human TNF was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT31 at 1 ug/mL was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. TNF band was visualized using ECL Substrate. Result: KT31 can detect recombinant human TNF by Western blotting.
