Cross reactivity test: Microtiter wells were coated with various human cytokines. K16225_7D3 was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: K16225_7D3 does not cross-react with human IL1B, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, IL17A, CSF2, IFNA, IFNG, CCL2, KITLG and TNF.
ELISA: Microtiter wells were coated with K16225_7D3 at 3 ug/mL as the capture antibody. Peroxidase conjugated mouse anti-human IL-2 monoclonal antibody (K16225_6D2) was used as the detection antibody. Result: K16225_7D3 and K16225_6D2 can be used as matched antibody pairs to detect and quantify the concentration of human IL-2.
Western Blot: 25 ng of recombinant human IL-2 was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K16225_7D3 at 1 ug/mL was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. IL-2 band was visualized using ECL Substrate. Result: K16225_7D3 can detect recombinant human IL-2 by Western blotting.
