Cross reactivity test: Microtiter wells were coated with human cytokines. K16225_2H10 was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. Results: K16225_2H10 does not cross-react with human IL1B, IL4, IL5, IL6, IL7, CXCL8, IL9, IL10, IL11, IL12, IL13, CSF2, IFNA, IFNG, CCL2, KITLG and TNF.
ELISA: Microtiter wells were coated with K16225_2H10 at 3.0 ug/mL as the capture antibody. IL2 (Cat. PP-323) was used as the antigen. Peroxidase conjugated mouse anti-human IL2 monoclonal antibody (K16225_3G1) was used as the detection antibody. Result: K16225_2H10 and K16225_3G1 can be used as a matched antibody pair to detect and quantify the concentration of IL2 .
Western Blot: 25 ng of recombinant human IL-2 was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose paper. K16225_2H10 at 1 ug/mL was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. IL-2 band was visualized using ECL Substrate. Result: K16225_2H10 can detect recombinant human IL-2 by Western blotting.
