ELISA: Microtiter wells were coated with K92011_3F10 at 3 ug/mL as the capture antibody. Synthetic peptide corresponding to amino acids 1-42 of human beta amyloid was used as the antigen. Peroxidase conjugated mouse anti-human beta amyloid 1-42 monoclonal antibody (K70013_6H1) was used as the detection antibody. Result: K92011_3F10 and K70013_6H1 can be used as a matched antibody pair to detect and quantify the concentration of human beta amyloid 1-42.
Western Blot: 25 ng of synthetic APP peptide fragments was crosslinked with BSA. The conjugate was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. K92011_3F10 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. APP band was visualized using ECL Substrate. Result: K92011_3F10 can detect APP by Western blotting.
