Cross reactivity test: Microtiter wells were coated with various peptides and recombinant proteins. KT232 was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: KT232 does cross-react with human MAPT.
ELISA: Microtiter wells were coated with KT234 at 3.0 ug/mL as the capture antibody. MAPT (Cat. PP-11309) was used as the antigen. Peroxidase conjugated mouse anti-human MAPT monoclonal antibody (KT232) was used as the detection antibody. Result: KT232 and KT234 can be used as a matched antibody pair to detect and quantify the concentration of MAPT .
Western Blot: 25 ng of recombinant human Tau protein was run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT232 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Tau band was visualized using ECL Substrate. Result: KT232 can detect recombinant human Tau by Western blotting.
