Cross reactivity test: Microtiter wells were coated with human immunoglobulins with various isotypes. 4C3 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: 4C3 does not cross-react with human IgG1, IgG2, IgG3, IgG4, IgA and IgM.
ELISA: Microtiter wells were coated with 4C3 at 3 ug/mL as the capture antibody. Human IgE was used as the antigen. Peroxidase conjugated mouse anti-human IgE monoclonal antibody (1A2) was used as the detection antibody. Result: 4C3 and 1A2 can be used as a matched antibody pair to detect and quantify the concentration of human IgE.
Western Blot: Various protein samples were run on 6-18% SDS-PAGE under reducing conditions and blotted onto PVDF membrane. 4C3 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Human IgE band was visualized using ECL Western Blotting Substrate. Lane 1: 100 ng of human IgG Lane 2: 100 ng of human IgA Lane 3: 100 ng of human IgM Lane 4: 100 ng of human IgE Result: 4C3 can detect human IgE by Western blotting.
