Cross reactivity test: Microtiter plates were coated with human immunoglobulins with various isotypes. KT209 was used as primary antibody and peroxidase conjugated goat anti-mouse IgG was used as secondary antibody. Results: KT209 does not cross-react with human IgG1, IgG2, IgG3, IgA, IgM and IgE.
ELISA: Microtiter wells were coated with KT209 at 5 ug/mL as the capture antibody. Human IgG4 was used as the antigen. Peroxidase conjugated mouse anti-human IgG4 monoclonal antibody (KT131) was used as the detection antibody. Result: KT209 and KT131 can be used as a matched antibody pair to detect and quantify the concentration of human IgG4.
Western Blot: Various protein samples were run on 6-18% SDS-PAGE under reducing conditions and blotted onto PVDF membrane. KT209 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. KT209 band was visualized using ECL Western Blotting Substrate. Lane 1: 100 ng of Human IgG1 Lane 2: 100 ng of Human IgG2 Lane 3: 100 ng of Human IgG3 Lane 4: 100 ng of Human IgG4 Lane 5: 100 ng of Human IgA Lane 6: 100 ng of Human IgE Lane 7: 100 ng of Human IgM Result: KT209 can detect human IgG4 by Western blotting.
