Cross reactivity test: Microtiter wells were coated with human immunoglobulins with various isotypes. KT132 was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Result: KT132 does not cross-react with human IgA, IgM and IgE.
ELISA: Microtiter wells were coated with KT46 at 3 ug/mL as the capture antibody. Recommbinant IGHG1 Protein (Cat. KC-001) was used as the antigen. Peroxidase conjugated Anti-Human IgG1 Antibody (KAA199_6A2) was used as the detection antibody. Result: KAA199_6A2 and KT46 can be used as a matched antibody pair to detect and quantify the concentration of IGHG1 .
Western Blot: Various protein samples were run on 6-18% SDS-PAGE under reducing conditions and blotted onto nitrocellulose membrane. KT132 at 1 ug/mL was used as the primary antibody and peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Human IgG band was visualized using ECL Western Blotting Substrate. Lane 1: 500 ng of human IgG1 Lane 2: 500 ng of human IgG2 Lane 3: 500 ng of human IgG3 Lane 4: 500 ng of human IgG4 Lane 5: 500 ng of human IgG Lane 6: 500 ng of human IgA Lane 7: 500 ng of human IgM Lane 8: 500 ng of human IgE Result: KT132 can detect human IgG by Western blotting.
